藏药新药研究与开发学科组在虎耳草属植物自由基抑制剂研究取得新进展
虎耳草科虎耳草属植物,民间以其全草入药,具有祛风、清热、凉血解毒等功效,传统用于治疗肝胆、风疹、湿疹、中耳炎、丹毒、咳嗽吐血、痔疾等疾病。中国科学院西北高原生物研究所藏药新药研究与开发学科组在前期研究中发现虎耳草属植物组分具有优异的体外清除自由基活性,但是并不清楚其所含的外源性自由基抑制剂物质基础。因此,学科组以DPPH自由基为靶标,以虎耳草属植物研究为例,构建了虎耳草属植物中自由基抑制剂的HPLC-DPPH在线筛选系统,是集在线筛选、靶向富集、定向纯化分离为一体的天然产物化学色谱研究体系。其次,基于HPLC-DPPH在线筛选系统,结合中压、高压色谱从黑虎耳草、黑蕊虎耳草和唐古特虎耳草植物中定向纯化分离并鉴定了51个自由基抑制剂,其中12个新化合物,1个新天然产物;最后,通过体外细胞实验阐明了唐古特虎耳草中3个新的二芳基壬烷类化合物Saxitanside A、Saxitansin B和Saxitansin D,具有抑制H2O2诱导的HepG2细胞氧化损伤的作用机制。
